Overwhelmed Ant McPartlin joked as he leaped on stage to collect the award from Kermit the Frog: "Again?"
Partner Dec Donnelly added: "We've been lucky enough to win a couple of these but every time it feels like the first time. We are just so honoured and humbled."
The pair have won the award every year since 2001 — except in 2009 when the awards were rescheduled to the following year.
Speaking later, Ant admitted: "We were more nervous this year than ever before. Ten years in a row was tremendous.
"But then, you get to the red carpet this year and think, 'I wouldn't mind another one. We're all dressed up!'"
The pair, who opened the awards with a song-and-dance routine to Let There Be Love with telly legend Bruce Forsyth, later also collected the Best Reality Show prize for I'm A Celebrity.
Thanking the Aussie-based series' staff, Dec told the audience: "All the crew make a big sacrifice being out in Australia, but nobody makes as much of a sacrifice as the male kangaroo population."
He also thanked last year's jungle campmates, saying: "Every year we get to Australia and people back home say, 'Oh the cast's rubbish, they're too old, or they're too this.
"And then you pull out a performance like these guys — a vintage 2011.
